Die ABCSG (Austrian Breast & Colorectal Cancer Study Group) ist ?sterreichs gr??te und bekannteste akademische Forschungsorganisation, die international erfolgreich klinische Studien zu Brust- und Darmkrebs durchführt. Seit 1984 haben rund 29.000 Patientinnen und Patienten weltweit an ABCSG-Studien teilgenommen. The Austrian Breast & Colorectal Cancer Study Group (ABCSG) is Austria’s biggest and best-established academic research organization that conducts internationally respected clinical trials to investigate breast and colon cancer. Our primary objectives are to standardise diagnosis, therapy and post-treatment of breast and colon cancer throughout Austria, and to provide patients with the latest and best possible treatments. More than 29,000 patients worldwide have participated in ABCSG trials since 1984.

?? A big step for #ABCSG 61 / TEODOR! We are thrilled to announce that the first site of our new national study, ABCSG 61 / TEODOR, has been activated! The Center for Breast Health in the Department of Gynecology at the Medical University of Vienna, headed by Prof. Christian Singer, is now ready to enroll patients. ABCSG 61 / TEODOR is a prospective, randomized, phase II trial that will investigate whether endocrine therapy is comparable to chemotherapy with regards to efficacy, while improving quality of life for patients with ER-positive, HER2-negative early and locally advanced breast cancer by using ctDNA and Ki-67 to guide treatment decisions. ?? ctDNA is assessed centrally with the Signatera? test by Natera. ?? Final survival Results from the PENELOPE-B trial published in Annals of Oncology ?? #Palbociclib fails to improve long-term survival in high-risk HR+/HER2? early breast cancer – but new hope for lobular breast cancer? The final analysis of the #PENELOPE-B trial provides critical long-term data on the efficacy of CDK4/6 inhibition in early high-risk hormone receptor-positive (HR+)/HER2-negative breast cancer with residual disease after neoadjuvant chemotherapy (#NACT). ?? Study Design & Purpose PENELOPE-B investigated whether the addition of 1 year of #palbociclib to standard adjuvant endocrine therapy (#ET) could improve outcomes for patients with high-risk HR+/HER2? breast cancer and residual invasive disease post-NACT. ?? Key Findings (Median follow-up: 77.8 months) No significant improvement in overall survival (OS) or invasive disease-free survival (iDFS) in the total study population. Results remained consistent across predefined subgroups. ?? Exploratory Insight: Lobular Breast Cancer (LBC) A glimmer of promise was observed in a post hoc analysis of patients with invasive lobular carcinoma (#ILC): #iDFS significantly improved: HR 0.52 (95% CI 0.28–0.97, P = 0.035) #OS showed a strong trend: HR 0.45 (95% CI 0.19–1.07, P = 0.062) ?? Conclusion While the overall results did not support routine use of palbociclib in the adjuvant setting for HR+/HER2? early breast cancer, these findings highlight the need for further research into histological subtypes such as lobular breast cancer, where CDK4/6 inhibition might offer more benefit. ??
